Reversed-Phase
Hamilton Reversed-Phase HPLC Columns
Hamilton’s reversed-phase HPLC columns are engineered to deliver exceptional inertness, longevity, and batch-to-batch consistency across a wide range of analytical workflows. By merging the strengths of both silica-based and polymeric stationary phases, these columns provide robust performance for laboratories seeking reliable separations of small molecules, peptides, pharmaceuticals, and complex organic mixtures.
Hamilton offers an extensive portfolio that includes four polymeric and two silica-based reversed-phase packing materials, each optimized for specific selectivity, retention, and chemical stability requirements. The polymeric phases provide outstanding resistance to high pH, aggressive mobile phases, and repeated cleaning cycles—ideal for high-throughput laboratories and challenging sample matrices. The silica-based phases deliver high efficiency and sharp peak shapes for traditional reversed-phase workflows, ensuring analytical precision even with demanding separations.
Together, these reversed-phase chemistries allow analysts to fine-tune methods for maximum resolution, reproducibility, and sensitivity. Whether you need rugged polymeric durability or classic silica-based selectivity, Hamilton’s HPLC columns offer reliable performance suitable for pharmaceutical, environmental, clinical, food and beverage, and research applications.

Hamilton HxSil reversed-phase columns are engineered with high-purity, well end-capped silica to deliver consistent retention, excellent reproducibility, and long-term durability in analytical HPLC workflows.
HxSil C18 columns provide enhanced hydrophobic retention compared to many conventional C18 phases, improving separation of compounds that are often poorly retained on standard columns. This makes HxSil C18 an ideal choice for challenging analytes requiring increased interaction with the stationary phase while maintaining reliable retention time stability.
HxSil C8 columns offer the same end-capped silica chemistry with reduced alkyl chain length, delivering excellent selectivity for complex sample mixtures while producing shorter retention times than C18. HxSil C8 is commonly selected when faster analyses are needed without sacrificing chromatographic resolution.
Key Performance Benefits
- High-purity, end-capped silica for reproducible retention
- Improved durability for routine analytical operation
- HxSil C18 for enhanced retention of weakly retained compounds
- HxSil C8 for faster separations with excellent selectivity
Both HxSil C18 and C8 phases are well suited for pharmaceutical, environmental, and research applications requiring dependable reversed-phase performance across a wide range of mobile phase conditions.

Hamilton PRP-1 Columns — Polymer Packing
Hamilton PRP-1 columns are designed to maximize sample recovery during purification and quantitation—especially for labile or reactive compounds that can irreversibly bind to silica-based C8/C18 phases. Because Hamilton PRP polymer supports are made entirely of poly(styrene-divinylbenzene) (PSDVB), they do not contain silanol groups that can cause analyte loss.
PRP-1 is particularly valuable for protected oligonucleotides and other sensitive analytes where recovery is critical. In many applications, PRP-1 can deliver 95% or greater recovery of the same sample that may only partially recover on traditional C18 columns—supporting higher confidence quantitation and improved workflow efficiency.
Unlike silica-based C8/C18 columns, PRP-1 has no stationary phase coating. With no coating to hydrolyze, the column maintains performance characteristics longer than many conventional reversed-phase columns, helping laboratories reduce downtime and replacement frequency.
Typical Analytes
Polycyclic aromatic hydrocarbons (PAH), ionizable organic compounds, steroids, and peptide fragments.
Applications
Well suited for small organic compounds (<2,000 MW), pharmaceutical analysis, steroid profiling, nucleic acid separations, vitamin analysis, and herbicide detection—especially when enhanced recovery and long-term stability are required.

Hamilton PRP-3 & PRP-h5 Columns — Polymer Packing
Hamilton PRP-3 is a polymeric reversed-phase HPLC column designed for the purification and isolation of proteins and peptides with excellent recovery (>90%). Built on an inert poly(styrene-divinylbenzene) (PSDVB) support, PRP-3 eliminates silanol-driven adsorption that can irreversibly bind biomolecules and reduce yield.
PRP-3 is based on PRP-1 chemistry but uses a larger 300 Å pore size (vs. PRP-1’s 100 Å), improving accessibility for larger analytes such as proteins, digests, and high molecular weight polymers.
PRP-h5 uses the PRP-3 base with a pentafluorinated modification, increasing hydrophobic character and delivering selectivity differences compared with standard silica C18 phases. This behavior can be especially useful for halogenated solutes and other analytes where altered selectivity improves separation.
Typical Analytes
PRP-3: Globular proteins, albumins, antibody fragments, tryptic digests, DNA, RNA oligomers, synthetic high MW polymers
PRP-h5: Oligonucleotides, angiotensin, apo-transferrin, apomyoglobin (equine), carbonic anhydrase, cytochrome C, myoglobin, ribonuclease A
Applications
PRP-3: Organic compounds and large molecules (>2,000 MW), peptides, proteins, protein digests, protected and de-protected oligonucleotides, nucleic acids
PRP-h5: Organic compounds and macromolecules (>2,000 MW), pharmaceuticals, protein digests, tryptic digests, and proteomics workflows where enhanced hydrophobicity and altered selectivity are beneficial
