AdvanceBio Glycan Mapping Amide HILIC
HILIC mode • Amide-bonded phase • Optimized for labeled glycans • Bio-inert hardware
â–¸

AdvanceBio Glycan Mapping Amide HILIC is specifically engineered for high-resolution separation of released and labeled glycans. The amide stationary phase supports hydrophilic interaction chromatography (HILIC), providing strong retention and excellent resolution of structurally similar N-linked and O-linked glycans.

When to Choose This Phase

  • 2-AB, 2-AA, RFMS, or other fluorescently labeled glycan analysis
  • High-resolution N-glycan profiling
  • Glycoform characterization of monoclonal antibodies
  • Biosimilar comparison and batch-to-batch QC testing

How It Works

In HILIC mode, glycans are retained based on hydrophilicity and hydrogen-bonding interactions. The amide functionality provides consistent retention and selectivity, enabling reliable separation of high-mannose, complex, hybrid, and sialylated glycan structures.

Key Definitions â–¸
HILIC (Hydrophilic Interaction Chromatography)
A chromatographic mode that retains highly polar compounds using a water-enriched layer on the stationary phase surface. In glycan analysis, HILIC enables strong retention and high-resolution separation of hydrophilic oligosaccharides.
Amide-Bonded Phase
A stationary phase functionalized with amide groups that provide consistent hydrogen bonding and polar interactions. In glycan mapping, this chemistry delivers reproducible retention and improved resolution of closely related glycan structures.
Released & Labeled Glycans
Glycans enzymatically cleaved from glycoproteins (commonly using PNGase F) and chemically labeled (e.g., 2-AB, 2-AA, RFMS) to enhance fluorescence or MS detection. This preparation enables accurate profiling and quantitation.
Glycan Mapping
The analytical process of separating, identifying, and quantifying glycan species attached to therapeutic proteins. Glycan mapping is critical for biopharmaceutical characterization, biosimilar comparison, and quality control.
Glycoform Profiling
The measurement of structural variants of glycans (high-mannose, hybrid, complex, sialylated, fucosylated, etc.) attached to a protein. Changes in glycoform distribution can affect therapeutic efficacy, stability, and immunogenicity.
Frequently Asked Questions â–¸
What is the AdvanceBio Glycan Mapping column used for?
AdvanceBio Glycan Mapping columns are designed for high-resolution separation of released and labeled glycans. They are commonly used for N-glycan profiling of monoclonal antibodies and other biotherapeutics in development and QC environments.
Why is an amide HILIC phase preferred for glycan analysis?
Glycans are highly polar molecules. The amide HILIC phase provides strong hydrophilic retention and excellent resolution of structurally similar glycan species, including high-mannose, hybrid, and complex glycans with varying degrees of sialylation and fucosylation.
Are these columns compatible with fluorescence and LC/MS detection?
Yes. AdvanceBio Glycan Mapping columns are fully compatible with fluorescence detection (FLD) for labeled glycans (such as 2-AB or 2-AA) and with LC/MS workflows when structural confirmation or mass analysis is required.
What types of glycans can be resolved using this column?
The column effectively separates high-mannose, hybrid, and complex glycans, including species with different levels of branching, galactosylation, fucosylation, and sialylation. It is optimized for subtle structural differences critical in biosimilar evaluation.
What applications benefit most from AdvanceBio Glycan Mapping columns?
These columns are widely used for biotherapeutic characterization, biosimilar comparison, batch release testing, stability studies, and regulatory submissions where precise glycan profiling is required.
