When performing HPLC, some substances are difficult to detect by traditional UV/VIS light absorption or florescence detectors. Often times these difficult-to-detect components get lost in the background noise. Post-column derivatization (or post-column reaction) works after the separation to chemically react with the substances to give them an easily detectable physical property. For example, a strong color or fluorescent product that can easily be detected against a complex background.
